The
eluents used were A: formic acid (0.5% v/v) and B: acetonitrile (LC-MS grade).
- Launching an HPLC Column to Analyze Anions, Compatible with Hydroxide
EluentsA good extraction solvent will contain all of the
eluents in the chromatography process so that any reactions of the sample resulting in precipitates will occur prior to injection into the HPLC.
The influence of organic modifier concentration, addition of salts, and pH of
eluents on chromatographic parameters such as retention, resolution, chromatographic plate numbers, and peak asymmetry was investigated.
Unfortunately,
eluents at pH value above 8 are not totally compatible with classic C18 or polymeric columns employed in chromatographic separations.
Thus, desorption studies were carried out using three different
eluents (neutral water, 0.1 M HCl and 0.1 M CH 3 COOH).
The
eluents used for the purification process were MeOH : [H.sub.2]O (50 : 50 v/v), CH[Cl.sub.3]: MeOH : [H.sub.2]O (22.5: 55 : 22.5 v/v), and CH[Cl.sub.3]: MeOH :[H.sub.2]O(70 : 26: 4 v/v).
Therefore, we just collected and combined the
eluents in the tubes within the same peak and dismissed others.

Reversed-phase HELC in
eluents of the acetonitrile-formic acid-water system was used in the isocratic modein
eluents from 6 to 10 vol.
